This article is a synthesis of the article "The Effect of Oral Administration of Bovine Bone Collagen Peptide on Skin Aging in Naturally Aging Mice" in the September 2017 issue of "Nutrients", Volume 9, Issue 1209, and the product features of Jiayi bovine bone collagen Peptide.
With people's pursuit of a youthful appearance, the demand for anti-aging intervention measures that delay or even reverse the signs of skin aging is increasing day by day. The use of dietary supplements to improve the appearance and function of aging skin has received increasing attention. Collagen peptides (CPs) are used as important active ingredients due to their various biological activities, high bioavailability and good biocompatibility. An international study has shown that oral administration of bovine bone collagen peptides can indeed alleviate natural skin aging and improve skin aging.
Experimental method
Experimental Animals and Grouping
13-month-old female Kunming mice (equivalent to 45 years old in humans) were divided into 7 groups according to body weight:
Young control group (Group Y, n=10) : 2-month-old mice were gavaged with normal saline;
Intervention group (n=50) : It is divided into ACP-200, ACP-400, ACP-800 (collagen peptides prepared by alkaline protease, with doses of 200, 400, and 800 mg/kg respectively), CCP-400 (collagen peptides prepared by collagenase, 400 mg/kg), and Pro-400 (proline, 400 mg/kg).
Method: All groups were intragastally administered once a day for 8 weeks.

Detection Indicators
Skin laxity (DSL) : Evaluated by measuring the stretched length of the skin on the back;
Skin histology: HE staining was used to observe the morphology of collagen fibers in the dermis;
Skin components: Determine the contents of hydroxyproline (Hyp), hyaluronic acid (HA), type I and type III collagen;
Antioxidant indicators: Detect the activities of superoxide dismutase (SOD), catalase (CAT), and the level of malondialdehyde (MDA).
Experimental results
The influence of collagen peptides on skin laxity
The skin laxity in the model group (Group M) was significantly higher than that in the young group (Group Y). After 8 weeks of intervention, the DSL in the ACP-400 group significantly decreased at the 6th week (p<0.05, p<0.05), and the DSL in the ACP-800 and CCP-400 groups decreased to the level of the younger group after 8 weeks. The DSL in the proline group (Pro-400) was also significantly improved (p<0.05p<0.05).

The Influence of collagen peptides on skin components
The hydroxyproline content in the ACP-800 group was significantly increased compared with the model group (p<0.05, p<0.05), and it was also significantly increased in the CCP-400 group.
Collagen ratio: The ratio of type I/type III collagen in the ACP-400 and ACP-800 groups returned to the level of the younger group (p<0.05, p<0.05), and the proline group also improved significantly;
Moisture and HA: There were no significant changes in skin moisture and HA content in each group.

Body weight, spleen index (SI) and Thymus index (TI)
During the experiment, the weight of the young group increased, while the weight of the model group remained stable. After the model group was treated with ACP (200, 400 and 800 mg/kg body weight), CCP and proline (400 mg/kg body weight) for 8 weeks, there was no statistically significant difference in body weight compared with the untreated model group. In addition, there were no significant differences in SI and TI between the ACP group, the CCP group and the proline group and the model group.
Skin Histology
In the model group (elderly mice), it was observed that the dermal tissue was lighter in color and had larger gaps. Compared with the young group, the dermis was thinner and there were fewer sebaceous glands. Compared with the model group, after the intake of ACP and CCP, the space in the dermal tissue decreased, and the fibers appeared denser and more organized. In addition, after treatment with ACP, the number of sebaceous glands increased, especially at a dose of 800 mg/kg body weight.
Skin Components
The skin moisture content and the ratio of type I collagen to type III collagen in the model group were significantly lower than those in the young group. Compared with the model group, the intake of CPs (including ACP and CCP) and proline had no significant effect on skin moisture and HA content. Conversely, oral ACP can increase the collagen content in a dose-dependent manner. There is a significant difference in collagen content between the ACP group with a body weight of 800 mg/kg and the model group (p<0.05). Oral administration of CCP at 400 mg/kg body weight could also increase the collagen content in the skin (p<0.05 compared with the model group). Among the groups with ACP intake, the ratio of type I collagen to type III collagen also showed a dose-dependent increase, and there were significant differences between the groups with ACP intake of 400 mg/kg and 800 mg/kg and the model group (all p<0.05).
Changes in antioxidant indicators
The activities of SOD and CAT in the skin of the model group were significantly lower than those of the young group. The MDA level in the model group was higher than that in the young group. Oral CPs (including ACP and CCP) significantly increased the activities of SOD and CAT and reduced the level of MDA (compared with the model group, all p<0.05). Furthermore, in the ACP administration group, the increase in the activities of SOD and CAT and the decrease in the level of MDA were dose-dependent.

This study shows that oral administration of bovine bone collagen peptides (CPs) can significantly improve skin laxity in naturally aging mice. The mechanisms include:
Collagen repair effect: CPs repairs the collagen fiber structure in the dermis by increasing the content of hydroxyproline and regulating the ratio of type I/type III collagen.
Antioxidant effect: CPs enhances the activities of SOD and CAT, reduces the level of MDA, and alleviates the damage of oxidative stress to the skin;
The difference in enzymatic hydrolysis methods: The CPs (ACP) prepared by alkaline protease is more effective than that prepared by collagenase (CCP), which may be related to the molecular weight distribution and the biological activity of the peptide segments.
Although proline can improve skin laxity, it does not show antioxidant activity, suggesting that the anti-aging effect of CPs involves multi-pathway synergy. This study provides experimental evidence for bovine bone collagen peptides as functional foods against skin aging.
